Research Applications
Compare reference anticancer agents and internally developed candidate inhibitors while accounting for model-specific culture and exposure conditions.
Model & Design
Four breast, three ovarian and two gastric cancer cell lines cultivated separately as low-attachment spheroids; five lines also evaluated as monolayers.
Experimental Format
Treatment began after reproducible aggregation. Spheroid exposure lasted 14 or 21 days by line; 2D exposure lasted 72 hours. Metabolic viability assays supported concentration–response analysis.
Model Components
Component
Breast models
Description
BT-20, HCC70, MDA-MB-453 and T47D cell lines.
Component
Ovarian and gastric models
Description
TOV21G, OVCAR-3 and A2780; SNU-1 and HGC-27. TOV21G required agarose under the selected conditions.
Component
Readouts
Description
AlamarBlue or PrestoBlue fluorescence normalized to matched untreated controls; fitted IC50 values when reliable and an integrated viability index as a supplementary response summary.
Measurements & Endpoints
Spheroid formation and morphology
Normalized metabolic viability
IC50 or reported range
Integrated viability index when curve fitting is insufficient
Analysis
Compare drug responses within each specified line and exposure schedule. Do not attribute a 2D–3D difference solely to geometry when the treatment duration also differs.
Published Evidence
Study Design
Nine cell-line spheroids tested with six reference agents, followed by candidate CDK7 inhibitors; five lines also assessed in 2D.
Study Sample
Nine cell lines in 3D; five of those in 2D. Candidate-inhibitor experiments were repeated at least twice with three technical replicates per concentration.
Comparison
Matched untreated wells within each model, reference agents, and 2D versus 3D conditions with different exposure durations.
Finding
Sensitivity changed by line and compound. Paclitaxel was a consistent assay control; iCDK7_1 showed relatively consistent antiproliferative activity in the tested spheroid panel.
Interpretation & Limits
2D exposure was 72 hours, whereas 3D exposure was 14 or 21 days.
The assay did not verify CDK7 target engagement in each model.
Compound stability over prolonged exposure was not directly measured.
Researchers optimized spheroids from nine breast, ovarian and gastric cancer cell lines before comparing anticancer compounds. Some 3D models appeared more sensitive than their 2D counterparts, while others appeared less sensitive; exposure times also differed.
