Research Applications

Identify medium-dependent pharmacology and assess whether response shifts reverse after medium transfer.

Model & Design

Patient-derived pancreatic and colorectal tumor organoids: three paired models in the drug screen and two models in transfer experiments.

Experimental Format

Paired Wnt/R-spondin/noggin and OncoPro cultures, stabilized for at least two passages before a 33-compound, four-point screen with imaging over five days.

Model Components

  • Component

    Patient-derived organoids and paired media

    Description

Tumor identity and expansion quality are checked separately; each condition stabilizes before screening.

  • Component

    Imaging and normalized analysis

    Description

Medium-matched vehicle and positive controls; normalized organoid growth rate and normalized area over the curve.

Measurements & Endpoints

  • Organoid growth and killing
  • Normalized drug-response curve area
  • Medium-associated transcriptional state
  • Response after medium transfer

Analysis

Compare media within each patient model. Report completed screens and expansion failures separately, and interpret medium effects for each compound and model.

Published Evidence

  • Study Design

Paired screening in two media followed by transfer experiments in two models.

    Study Sample

Three patient models and 33 compounds; two technical and two biological replicates per patient. Biological replicates were wells cultured separately for at least two passages.

    Comparison

Each model in Wnt and OP, with controls matched to its medium.

    Finding

Some targeted agents were more active in Wnt; platinum curves were largely flat over the tested range; selected sensitivity shifts were reversible.

    Interpretation & Limits
    • The pharmacology sample was small and was not compared with patient treatment outcomes.
    • Medium effects varied by drug and model; they cannot be generalized to all tumors or treatment classes.
